실 한가닥을 마구 뭉쳤다가 양쪽 끝을 잡고 당기면 당연하게도 매듭(knot)이 생긴다. 단백질 같은 작은 끈에서는 매듭이 우리 신발끈보다도 더 잘 생긴다.

그런데 실제 존재하는 단백질에는 매듭이 매우 드물다. 생명체에서 발견되는 모든 단백질 중에 1%정도만이 매듭 구조를 가지고 있으며, 그마저도 가장 단순한 매듭들이다. 그리고 이런 매듭을 가진 단백질들은 모두 효소로 쓰인다고 한다. Virnau씨는 새로운 알고리즘을 개발해서 PDB에 있는 단백질들이 가진 매듭을 찾아보았는데, 지금까지 발견되지 않았던 가장 복잡한 매듭을 찾았다고 한다1.

재미있는 건, 가장 복잡한 매듭을 가진 그 단백질이 하는 일이 매우 특별하다는 것이다. 그 단백질의 이름은 "ubiquitin hydrolase"인데, 단백질의 사망예고딱지인 ubiquitin을 분해하여 파괴되기 직전의 단백질을 구출하는 역할을 한다. 이 단백질이 이런 일을 할 수 있는 이유에 대해서 연구팀은 단백질을 분해하는 proteasome이 이 매듭을 잘 풀지 못해서가 아닐까라고 추측한다고 한다.

Ubiquitin은 어디에나(ubiquitous) 존재하는, '죽음이라는 숙명'이다. 하지만 저 복잡한 매듭을 가진 단백질은 그 풀 수 없는 매듭 덕에 이러한 운명을 좌지우지 할 수 있는 특별한 존재가 된 것이다.
